What Is Direct DIA?
Direct DIA (direct Data Independent Acquisition) is an evolved DIA based quantitative proteomics strategy that identifies and quantifies peptides directly from DIA data without requiring a pre built DDA spectral library. Traditional DIA normally depends on an external spectral library built from DDA data. This adds time, cost, and sample consumption, and can limit coverage if the library does not fully match the study matrix or conditions. Direct DIA removes this dependency.
Instead of relying on separate DDA runs to construct a library, Direct DIA uses in silico predicted spectra, on the fly pseudo libraries, or DIA data itself to perform peptide centric identification and quantification.
